Project Title: Supportive activities for the Tuberculosis study in the Dar es Salaam Region

Project Description: Our overall goal is to initiate a TB cohort of sputum smear-positive HIV-infected and HIV-negative TB cases in the metropolitan setting of the large African city Dar es Salaam with a high TB incidence. Specifically, we aim: • to collect TB key variables to study the TB epidemiology, including data on TB/HIV co-treatment, TB treatment outcomes, frequency of diabetes, previous history of TB and recurrent TB, frequency of additional extrapulmonary manifestations, geographical information of the... Our overall goal is to initiate a TB cohort of sputum smear-positive HIV-infected and HIV-negative TB cases in the metropolitan setting of the large African city Dar es Salaam with a high TB incidence. Specifically, we aim: • to collect TB key variables to study the TB epidemiology, including data on TB/HIV co-treatment, TB treatment outcomes, frequency of diabetes, previous history of TB and recurrent TB, frequency of additional extrapulmonary manifestations, geographical information of the patients’ household; to identify risk factors for treatment failures and death, and risk factors for additional extra-pulmonary manifestations (25); • to collect and store the sputum samples and Mtb culture isolates from the recruited TB patients for further molecular investigations, such as standard molecular genotyping, phylogenetic markers and whole genome sequencing (11;12); • to collect and store blood, urine and stool samples from TB cases at baseline and during follow-up as well as from a control population without TB from the TB household contacts to evaluate new TB diagnostics, biomarkers, parasitic co-infections, and the nutritional status (26); • to provide baseline data of the TB study population in the Dar es Salaam region, and to prepare the field for future TB projects embedded in this cohort project.
